Bile Acid Synthesis Disorders in Japan: Long-Term Outcome and Chenodeoxycholic Acid Treatment

چکیده
We encountered 7 Japanese patients with bile acid synthesis disorders (BASD) including 3?-hydroxy-?5-C27-steroid dehydrogenase/isomerase (3?-HSD) deficiency (n = 3), ?4-3-oxosteroid 5?-reductase (5?-reductase) and oxysterol 7?-hydroxylase 1) over 21 years between 1996 2017. aimed to clarify long-term outcome in the BASD as well efficacy of chenodeoxycholic (CDCA) treatment 5 3?-HSD or deficiency. Diagnoses were made from genetic analyses. Bile analysis serum urine was performed using gas chromatography–mass spectrometry. Clinical laboratory findings profiles at diagnosis most recent visit retrospectively obtained medical records. Long-term included follow-up duration, treatments, growth, education/employment, complications treatment, other problems. Medians ranges current patient ages duration CDCA are 10 (8 43) 21), respectively. All patients, who had homozygous compound heterozygous mutations HSD3B7, SRD5B1, CYP7B1 gene, currently good health without liver dysfunction. In hepatic function gradually improved following initiation. No adverse effects noted. concluded that is effective deficiency, cholic has been countries. carry a prognosis early initiation treatment.

Cyclosporin A blocks bile acid synthesis in cultured hepatocytes by specific inhibition of chenodeoxycholic acid synthesis.
Bile acid synthesis, determined by conversion of [4-14C]cholesterol into bile acids in rat and human hepatocytes and by measurement of mass production of bile acids in rat hepatocytes, was dose-dependently decreased by cyclosporin A, with 52% (rat) and 45% (human) inhibition of 10 microM. The decreased bile acid production in rat hepatocytes was due only to a fall in the synthesis of beta-muric...
متن کاملUrsodeoxycholic acid, chenodeoxycholic acid, and 7-ketolithocholic acid are primary bile acids of the guinea pig.
Guinea pig gallbladder bile contains chenodeoxycholic acid (62 +/- 5%), ursodeoxycholic acid (8 +/- 5%), and 7-ketolithocholic acid (30 +/- 5%). All three bile acids became labeled to the same specific activity within 30 min after [3H]cholesterol was injected into bile fistula guinea pigs. When a mixture of [3H]ursodeoxycholic acid and [14C]chenodeoxycholic acid was infused into another bile fi...

We have prepared a new panel of 23 BA derivatives of DCA, chenodeoxycholic acid (CDCA) and lithocholic acid (LCA) in order to study the effect of dual substitution with 3-azido and 24-amidation, features individually associated with cytotoxicity in our previous work. The effect of the compounds on cell viability of HT-1080 and Caco-2 was studied using the 3-[4,5-dimethylthizol-2-yl]-2,5-dipheny...
متن کاملPregnancy and bile acid disorders.
During pregnancy, extensive adaptations in maternal metabolic and immunological physiology occur. Consequently, preexisting disease may be exacerbated or attenuated, and new disease susceptibility may be unmasked. Cholestatic diseases, characterized by a supraphysiological raise in bile acid levels, require careful monitoring during pregnancy. This review describes the latest advances in the kn...
متن کاملMucin in gall bladder bile of gall stone patients: influence of treatment with chenodeoxycholic acid and ursodeoxycholic acid.
The concentration of hexosamine, a marker for mucin, was determined and related to the degree of cholesterol saturation and to the occurrence of cholesterol crystals in gall bladder bile of gall stone patients (n = 40) and gall stone free subjects (n = 25). Ten of the gall stone patients had been treated with chenodeoxycholic acid (CDCA) and eight with ursodeoxycholic acid (UDCA) three to four ...
